19781.接种耐盐植物乳杆菌对不同盐度发酵辣椒挥发性成分的影响

用顶空固相微萃取(SPME)的方法预处理盐渍辣椒样品,结合气相色谱—质谱联用仪(GC—MS),分析未接种和已接种耐盐优势菌在10%和20%盐度盐渍辣椒的挥发性物质,四组样品共检出194 种挥发性物质,共有物质种27 种。种类数:已接种20%盐度辣椒(94 种)>已接种10%盐度辣椒(92 种)>未接种10%盐度辣椒(83 种)>未接种20%盐度辣椒(78 种);挥发性物质总含量:已接种20%盐度辣椒(75.63%)>已接种10%盐度辣椒(66.89%)>未接种10%盐度辣椒(66.43%)>未接种20%盐度辣椒(53.31%)。盐渍辣椒中的挥发性香气物质主要包括醇类、酸类、酯类、醛酮类和烷烃类等众多复杂的香气成分,接种耐盐优势菌种后挥发物质种类、含量明显增加多,有助于盐渍辣椒香味物质形成。

19782.鳕鱼骨活性钙的制备及其生物利用度研究

目的:以水产加工下脚料鳕鱼骨为原料,探讨鳕鱼骨活性钙的最佳制备条件,并对其生物利用度进行研究。方法:对鱼骨进行基本组成分析与组织学观察。采用正交实验确定鱼骨活性钙的最佳提取条件。采用低钙饮食建立大鼠缺钙模型,检测大鼠血清生化指标、骨钙含量及钙代谢指标,对鱼骨活性钙的生物利用度进行探讨。结果:鳕鱼骨中Ca 与P 的含量分别达到20.78%和10.09%,其比值接近2:1。扫描电镜观察发现鳕鱼骨表面具有坚固致密的扁平板状羟基磷灰石结晶。采用正交实验确定活性钙最佳提取条件为:提取温度100℃,提取时间120min,酸混合比例1:2,酸质量分数15%,在此条件下提取率达到96.15%。与空白对照组相比,活性钙各剂量组能显著提高大鼠骨钙含量。钙代谢实验测定结果显示活性钙高剂量组钙吸收率显著高于相同钙水平的碳酸钙对照组。结论:鳕鱼骨可作为一种优良的天然钙源,鱼骨活性钙能够作为一种有效的钙营养强化剂加以开发。

19783.葡聚糖修饰米根霉脂肪酶条件优化及其稳定性

采用经高碘酸钠活化的葡聚糖修饰米根霉脂肪酶,以获得较高酸解催化活性。研究反应时间、反应pH、还原剂浓度及葡聚糖浓度对化学修饰效果的影响,并利用Box-Behnken 设计法及响应面优化法对米根霉脂肪酶的化学修饰条件进行优化,当pH 7.63,时间为15.53 h,还原剂浓度为0.20 mol/L,葡聚糖浓度为0.30 %时,脂肪酶酸解率可达到最高值37.28 %。在40℃最适温度下,修饰酶的活性比原酶提高2.2 倍,化学修饰后脂肪酶的稳定性有显著提高。

采用硫酸铵分级沉淀,CM-sepharose 离子交换层析,Superdex-200 凝胶过滤层析方法,从芫荽中分离纯化出电泳纯的酸性磷酸酶(acid phosphatase,ACP)。该酶的回收率为14.2%,纯化倍数为238.6 倍,比活力为295.87U/mg,亚基分子质量约为53.8kD;ACP 酶学性质研究表明:最适反应温度为55℃,在温度为40℃以下时较稳定,因此该酶对温度较敏感;最适反应pH 为5.8,在pH4-7 之间较稳定,表明该酶耐受于酸性环境;ACP 对对硝基苯酚的Km 值为0.63mmol/L,表明该酶与底物对硝基苯酚具有较高的亲和力;甲醇、乙醇、异丙醇、抗坏血酸、草酸、Cu2+、Pb2+、Ag+对该酶具有强烈的抑制作用;Mg2+、Mn2+、Ba2+、K+对该酶具有一定的激活作用。
