The purpose of this project is to develop DNA aptamer complexes that are selectively cytotoxic to PSMA+ prostate cancer (PCa) cells. Our studies showed Zn2+ is cytotoxic to prostate cancer cells and also sensitizes PCa cells to chemotherapy. We developed a new DNA motif for Zn2+ delivery. We also developed a new chemical strategy for delivering the cytotoxic drug doxorubicin to cancer cells. We have demonstrated that our novel dimeric aptamer complexes are selectively internalized by PSMA+ prostate cancer cells and have demonstrated selective cytotoxicity with delivery of doxorubicin. We also developed a new nanoparticle (NP) composed of porphyrin and DNA and demonstrated that this NP was cytotoxic to bladder cancer cells in vitro, was non-toxic in vivo, and displayed strong anti-tumor activity in vivo. The aptamer complexes and the NPs we developed in this project are highly selective and highly active agents that have the potential to markedly improve chemotherapy for treatment of advanced prostate cancer.

The electric power system in North America is linked between the United States and Canada. Canada has historically been a net exporter of electricity to the United States. The extent to which this remains true will depend on the future evolution of power markets, technology deployment, and policies. To evaluate these and related questions, we modify the Regional Energy Deployment System (ReEDS) model to include an explicit representation of the grid-connected power system in Canada to the continental United States. ReEDS is unique among long-term capacity expansion models for its high spatial resolution and statistical treatment of the impact of variable renewable generation on capacity planning and dispatch. These unique traits are extended to new Canadian regions.

Ductal carcinoma in situ (DCIS) makes up 18of all new breast cancer diagnoses, and is considered a precursor to invasive breast cancer even though the majority of cases almost 70may never progress to invasive disease. Markers that identify which patients are most likely to experience progression are critically needed so that fewer patients are over-treated. This study is evaluating two novel tumor markers that may indicate greater risk of tumor progression based on recent work that suggests that stromal syndecan-1 expression induces an extracellular matrix with aligned collagen fiber architecture, and that this collagen alignment in turn facilitates malignant cell invasion. We are using archived tumor tissue from 267 cases of DCIS of the breast to evaluate syndecan-1 expression and collagen alignment. These DCIS cases, diagnosed between 1995 and 1999, have been followed for breast cancer outcomes; to-date, 13of cases have experienced a second breast cancer diagnosis. Analysis of syndecan-1 expression and collagen alignment patterns will be completed soon, and preliminary data analysis has begun. Findings from this research are expected to be disseminated through peer-reviewed journals and for presentation at national conferences by the end of the year.

3414.专门针对卵巢癌与叶酸结合的纳铈的临床前评估

Despite the fact that there have been significant developments in anti-cancer technology, ovarian cancer still remains one of the leading causes of gynecological cancer deaths in the United States. To contribute towards novel approaches with minimal toxicity in treatment of ovarian cancer we proposed the present work, where we are integrating the field of nanotechnology with ovarian cancer cells unique property of overexpressing folic acid receptor alpha (FR- ) to specifically target ovarian cancer. A cerium oxide nanoparticle, called Nanoceria (NCe), that has the ability to act as an anti- oxidant in similar capacity as biological anti-oxidants was selected. NCe in our hands exhibited efficacy in inhibiting ovarian tumor growth in vivo. Taking advantage of the nanotechnology, NCe was conjugated with molecules of folic acid to specifically target ovarian tumors as they are known to over-express folate receptor. This would result in specific killing of ovarian cells by NCe while minimizing toxicity to normal tissue. Dr Seal s laboratory (UCF) was able to successfully conjugate NCe to folic acid to prepare folic acid tagged Nanoceria called FA-NCe. FA-NCe was tested in vitro and was observed to have higher anti-growth activity against ovarian cancer cells. When given in animals, FA-NCe was more potent in inhibiting ovarian tumor growth than NCe alone.

Wireless sensor networks are widely deployed in various areas of our daily life.The lifetime of wireless sensor networks takes significant effect in the performance of the whole network.Many network protocols are proposed to improve the lifetime of networks,and among them,low-energy adaptive clustering hierarchy (LEACH) is a promising protocol with self-organization of the clusters and periodic rotation of cluster heads selection to distribute energy consumption uniformly.LEACH performs well compared with other protocols on lifetime while it is simplified and has aspects to improve.In this paper,we modified the procedure of cluster heads selection in LEACH and proposed a protocol LEACH-CHE with consideration on the energy consumption of cluster heads.We analyzed LEACH-CHE and with the results of simulations,LEACH-CHE over performs on network lifetime compared with the original LEACH.

In order to solve the problem which is very common when we use a solar water heater for a shower but the hot water can not come immediately because of the cooled water remained in the pipeline, a no-pump cycling water-saving equipment is designed based on a pressure-regulated suction device, temperature acquisition control circuit and a water-saving box.The kinematics and mechanics parameters are confirmed on the result of calculation and software simulation, and are tested by doing physical experiment.It can make it come true to economize the water and energy for solar power heater.

3417.预防骨折的新的治疗策略

Falls and debilitating bone fractures are a major problem for veterans, and more than 40,000 veterans suffered hip fractures from 2000-2002. Falls are the main etiological factor in more than 90of fractures, and so treatments that can improve muscle strength while at the same time increasing bone mass will significantly reduce fracture-related morbidity and mortality. Myostatin is a factor that induces muscle wasting and suppresses bone formation. Our data collected thus far demonstrate: (i) myostatin suppresses proliferation of aged, but not young, myoblasts, (ii) myostatin is elevated with age in muscles composed primarily of slow-twitch fibers (e.g. soleus), and (iii) myostatin increases muscle mass and muscle fiber size in aged mice. These findings suggest that myostatin inhibitors may have potential for suppressing muscle wasting and improving muscle repair in older individuals, but their effect on bone may be less significant.

Moore’s law has allowed the microprocessor market to innovate at an astonishing rate. We believe microchip implants are the next frontier for the integrated circuit industry. Current health monitoring technologies are large, expensive, and consume significant power. By miniaturizing and reducing power, monitoring equipment can be implanted into the body and allow 24/7 health monitoring. We plan to implement a new transmitter topology, compressed sensing, which can be used for wireless communications with microchip implants. This paper focuses on the ADC used in the compressed sensing signal chain. Using the Cadence suite of tools and a 32/28nm process, we produced simulations of our compressed sensing Analog to Digital Converter to feed into a Digital Compression circuit. Our results indicate that a 12-bit, 20Ksample, 9.8nW Successive Approximation ADC is possible for diagnostic resolution (10 bits). By incorporating a hybrid-C2C DAC with differential floating voltage shields, it is possible to obtain 9.7 ENOB. Thus, we recommend this ADC for use in compressed sensing for biomedical purposes. Not only will it be useful in digital compressed sensing, but this can also be repurposed for use in analog compressed sensing.

In this paper,we discuss a method for implementing online Viterbi algorithm as standard relational queries over streaming data.Our primary contribution is an approach to storing and manipulating most probable state paths,the candidate solutions to the problem,within a relational data model.We evaluate our prototype implementation over real-world sensor data from the Intel Lab dataset.
