  • 4792.随机二期试验的模拟肽疫苗佐剂WT-1恶性胸膜间皮瘤患者完成后的多学科治疗

    The Wilms' tumor gene, WT1, encodes transcription factors that regulate cell proliferation, differentiation, and apoptosis. WT1 protein is highly expressed in malignant pleural mesothelioma (MPM), and is a rational target for immunotherapy. We have developed a vaccine comprised of four WT1 heteroclitic peptides that are given together with Montanide and GM-CSF as immunologic adjuvants. This WT1 vaccine was previously tested in a small pilot trial, and shown to be safe and immunogenic. We have chosen to test the efficacy of this vaccine in MPM patients who have minimal disease burden after completion of multimodality therapy, but remain at exceedingly high risk for recurrence. The specific aim of this project is to conduct a multicenter, blinded, randomized trial comparing treatment with the WT-1 peptide vaccine + Montanide/GM-CSF to treatment with Montanide/GM-CSF alone in patients with MPM who have completed multimodality therapy. The primary endpoint is progression free survival. The trial has opened at Memorial Sloan-Kettering and is actively enrolling patients.

    A key mission of the U.S. Department of Energy (DOE) Office of Electricity Delivery and Energy Reliability (OE) is conducting research and development to enhance the security and reliability of the nation's energy infrastructure. Improving the security of control systems, which enable the automated control of our energy production and distribution, is critical for protecting the energy infrastructure and the integral function that it serves. The DOE-OE Control Systems Security Program is actively pursuing advanced security solutions for control systems. The focus of this report is analyzing how, where, and what type of wireless communications are suitable for deployment in the electric power system and to inform implementers of their options in wireless technologies. The discussions in this report are applicable to enhancing both the communications infrastructure of the current electric power system and new smart system deployments.

    The current scarcity of public charging infrastructure is one of the major barriers to mass household adoption of plug-in electric vehicles (PEVs). Although most PEV drivers can recharge their vehicles at home, the limited driving range of the vehicles restricts their usefulness for long-distance travel. In this paper, an agent-based information system is presented for identifying patterns in residential PEV ownership and driving activities to enable strategic deployment of new charging infrastructure. Driver agents consider their own driving activities within the simulated environment, in addition to the presence of charging stations and the vehicle ownership of others in their social network, when purchasing a new vehicle. Aside from conventional vehicles, drivers may select among multiple electric alternatives, including two PEV options. The Chicagoland area is used as a case study to demonstrate the different deployment scenarios are analyzed.

    This thesis presents the development and evaluation of a distributed agent based system using reputation based trust and game theoretic techniques to improve the defense of the future smart grid from cyber-attack and equipment malfunctions. Future smart grid capabilities promise to leverage network technologies to revolutionize the production, transmission, distribution and consumption of electrical power. However, the internet like communication also increase the power grid's vulnerability to cyber-attack. This thesis uses simulation linking power systems with communication networks to demonstrate the benefits of a Distributed Decision Making Communication Enable Special Protection System (SPS) using reputation based trust and game theory to protect the power grid from malicious and non-malicious malfunctions. The simulations show that a distributed approach to SPS load shedding successfully maintains power grid stability after an electrical disturbance while using reputation based trust to defend the load shedding from cyber-attack and equipment malfunction. Additional simulations demonstrate the application of game theory to defend the SPS load shedding process when available resources prevent the monitoring and defense of every part of the power grid. The demonstrated capability increases the resiliency of the power grid by preventing uncontrolled blackouts through detection and mitigation of attacks, improving the system's reliability.

    In the United States, annual vaccination against seasonal influenza is recommended for all persons older than 26 months. Each season since 2004-05, CDC has estimated the effectiveness of seasonal influenza vaccine to prevent influenza-associated, medically attended acute respiratory infection (ARI). This season, early data from 1,155 children and adults with ARI enrolled during December 3, 2012-January 2, 2013 were used to estimate the overall effectiveness of seasonal influenza vaccine for preventing laboratory-confirmed influenza virus infection associated with medically attended ARI. After adjustment for study site, but not for other factors, the estimated vaccine effectiveness (VE) was 62(95confidence intervals (CIs) = 51-71). This interim estimate indicates moderate effectiveness, and is similar to a summary VE estimate from a meta-analysis of randomized controlled clinical trial data; final estimates likely will differ slightly. As of January 11, 2013, 24 states and New York City were reporting high levels of influenza-like illness, 16 states were reporting moderate levels, five states were reporting low levels, and one state was reporting minimal levels. CDC and the Advisory Committee on Immunization Practices routinely recommend that annual influenza vaccination efforts continue as long as influenza viruses are circulating. Persons older than 6 months who have not yet been vaccinated this season should be vaccinated. However, these early VE estimates underscore that some vaccinated persons will become infected with influenza; therefore, antiviral medications should be used as recommended for treatment in patients, regardless of vaccination status. In addition, these results highlight the importance of continued efforts to develop more effective vaccines.

These sensors measure important biogeochemical parameters for long deployments, enabling the capture of data at time scales over which they vary most meaningfully. The high-frequency, real-time water-quality data they generate provide opportuni-ties for early warning of water-quality deterioration, trend detection, and science-based decision support.

    In this paper we give a general overview of the development process carried out in the Desire project. This process can be roughly split up into four main phases, from specification in the beginning over two integration phases up to optimization in the end. With a large variety of technologies and expertise involved, the development of a complex service robot system such as the DESIRE technology platform proved to reveal many challenges on technical as well as organizational level. Several of these issues are highlighted in this paper with the goal to derive useful guidelines for other robotics projects.

    The report contains the results of a test series to evaluate the electrical environment of a motor vehicle during normal operating conditions and a summary of power supply variations and electrical signal transient characteristics. Both source and coupled signals were measured. A computerized coupling analysis program was used to determine the effects of body shielding,aperture size,and cable lengths on signal coupling in the 100to 200MHz band between a simulated mobile radio emission and a modeled air-cushion restraint system cable as it might be used in a motor vehicle. A series of susceptibility tests were performed on an electronic speed control system and an anti-skid control module to determine functional upset levels of injected signals at critical circuit ports on these devices. The upset criteria were based on performance departures from normal,resulting from the injection of interfering signals. The injected signals were designed to represent levels and durations characteristic of those generated within the vehicle or coupled from external sources.

  • 4800.TPD52:一种新型的前列腺癌疫苗

    Tumor protein D52 (D52) is a novel self-onco-antigen involved in cellular transformation, proliferation and metastasis that is over-expressed in prostate cancer cells. The overall goal of this Award is to test the efficacy of D52-based vaccines in the TRAMP murine model of prostate cancer, and to characterize vaccine induced mechanisms of tumor immunity. Due to unforeseen circumstances during this funding period primarily involving the animal vendor and maternity leave for funded personnel we lost over 6 months of productivity. Consequently, we requested and were granted a no cost extension through 8/31/2012. Despite this unfortunate loss of time, over the past 12 months we made the following significant findings: Heterologous prime-boost vaccination induced more than 80protection from primary tumor challenge the best we have observed thus far, but only 30protection from secondary challenge about 8 months later. Similarly, if animals were immunized and rested for 8 months before challenge only about 50were protected from significant tumor growth suggesting that induction of a more durable response may require modulation of peripheral mechanisms of immune suppression as we previously reported. Finally, we demonstrated for the first time in vivo the critical importance of both CD4+ and CD8+ effector T cells in tumor protection following D52 immunization.
