    Mental disorders among children are described as serious deviations from expected cognitive, social, and emotional development (US Department of Health and Human Services Health Resources and Services Administration, Maternal and Child Health Bureau. Mental health: A report of the Surgeon General. Rockville, MD: US Department of Health and Human Services, Substance Abuse and Mental Health Services Administration, Center for Mental Health Services, and National Institutes of Health, National Institute of Mental Health; 1999). These disorders are an important public health issue in the United States because of their prevalence, early onset, and impact on the child, family, and community, with an estimated total annual cost of $247 billion. A total of 13%20% of children living in the United States experience a mental disorder in a given year, and surveillance during 19942011 has shown the prevalence of these conditions to be increasing. Suicide, which can result from the interaction of mental disorders and other factors, was the second leading cause of death among children aged 1217 years in 2010. Surveillance efforts are critical for documenting the impact of mental disorders and for informing policy, prevention, and resource allocation. This report summarizes information about ongoing federal surveillance systems that can provide estimates of the prevalence of mental disorders and indicators of mental health among children living in the United States, presents estimates of childhood mental disorders and indicators from these systems during 20052011, explains limitations, and identifies gaps in information while presenting strategies to bridge those gaps.

    Mining communities access to appropriate health care has not been well studied. Appropriate care includes competencies to adequately diagnose and treat mining related disease or illness. The current study has two primary objectives. First, we seek to understand whether mining communities face shortages in numbers of safety net providers compared to non-mining communities. To accomplish this, the study locates and maps mine locations in the U.S. and rural safety net providers (Rural Health Clinics, Federally Qualified Health Centers, Critical Access Hospitals and Black Lung Clinics). Second, we assess the capacity of rural safety net providers in mining areas to prevent, diagnose and treat mining related injury and illness. This was done through a survey of rural primary care providers practicing in safety net facilities co-located with mining communities.

  • 1214.金属化DNA寡核苷酸适配子对于前列腺癌的治疗

    The purpose of this project is to develop aptamer-targeted DNA:Zn2+ complexes that are highly efficacious for prostate cancer treatment. Significant progress has been made on refining novel Zn2+-binding DNA motifs that utilize FdU nucleotides and that are highly cytotoxic towards prostate cancer cells. We demonstrated netropsin binding to the 3 -FdU DNA hairpin, clarified the mode of Zn2+ binding to this complex and demonstrated the Zn2+/DNA/netropsin ternary complex is highly cytotoxic towards prostate cancer cells. We also demonstrated DNA hairpins with stems consisting of G-FdU base pairs form complexes with Zn2+ that have increased stability at physiological pH relative to stems consisting of A-FdU base pairs. We completed SELEX and identified a new DNA aptamer to PSMA and demonstrated selective binding towards PSMA+ prostate cancer cells. We demonstrated that Zn2+-pyrithione is synergistic with FdUMP 10 for inducing cytotoxicity towards prostate cancer cells. We have purchased animals and are ready to evaluate Zn2+- pyrithione/FdUMP 10 in vivo in the coming months. All components are in place for forming the dimeric aptamer complexes and evaluating cytotoxicity in vitro and in vivo.

    NORC at the University of Chicago is pleased to submit this final report for Assessing the Status and Prospects of State and Local Health Department Information Technology Infrastructure, a project of the Assistant Secretary for Planning and Evaluation (ASPE) at the United States Department of Health and Human Services. NORC and ASPE worked in close partnership with the National Association of County and City Health Officials (NACCHO) and the Association of State and Territorial Health Officials (ASTHO). This report describes how state health agencies (SHAs) and local health departments (LHDs) use information technology systems to capture, manage, analyze, and report information. We assess trends in use of information technology (IT) among SHAs and LHDs following the Health Information Technology for Economic and Clinical Health (HITECH) Act of 2009, the Patient Protection and Affordable Care Act (ACA) of 2010, recent work in public health informatics, and other factors. This project assumes that IT tools and resources should play a role in governmental public health practice. However, we also assume that rigorous assessments of public health goals should motivate IT projects and that stakeholders should design and implement IT in direct service of those goals.

    Increased use of public transportation is an effective means of decreasing roadway congestion and its associated externalities. To increase the use of public transportation under economic and resource constraints, it is important to improve the understanding of public transportation supply characteristics and demand behavior and make use of this understanding to improve planning and operations functions. Analyzing and interpreting in situ public transportation conditions that are readily accessible and observable can greatly improve this understanding. Project investigators previously worked with The Ohio State University (OSU) Campus Area Bus Service (CABS) and a private technology provider to equip the CABS network with state-of-the-art sensing, communications, and passenger information systems that are presently used to provide realtime bus arrival information to CABS users and ridership and location information to CABS operators and planners.

    The purpose of the overall protocol is to study the metabolism and protein expression in the urine and blood of human subjects administered hydrocodone. An opioid is prescribed as a pain medication to the patient to minimize pain. Hydrocodone will be administered to healthy volunteers. Urine and blood will be collected prior to and following administration of the drug. The three separate biofiuids will be analyzed for drug and metabolites and for changes in protein expression. Changes in protein expression will provide a general understanding of opioid exposure in future studies relating to opioid abuse.

    Hemostasis is arguably the highest treatment priority for the polytraumatized patient. Best hemostasis practices cannot be developed if we lack a more full understanding of the cell-based clotting system during trauma. In order to address this fundamental knowledge gap, this study will provide a comprehensive characterization of the most important cellular component of the coagulation system, the platelet. The study proposes a comprehensive characterization of platelet function over time in civilian victims of polytrauma that may inform development of new diagnostic, therapeutic, and clinical guidelines for hemorrhage control and treatment of coagulopathy in trauma. We have successfully developed a comprehensive panel of tests to include: 1. Platelet aggregation by aggregometry under conditions of low and high shear stress and in response to collagen and ADP stimulation, 2. Platelet- associated thrombin generation using calibrated automated thrombography, 3. Platelet-induced clot contraction and using viscoelastic measures such as TEG with Platelet Mapping and, 4. Flow cytometry for platelet receptor and white cell activation as well as circulating platelet and platelet-monocyte aggregates. These panels are being coupled with a comprehensive data base of trauma victim demographics, diagnostics, and therapeutics to define the relationships between platelet function and injury pattern/severity, tissue hypoperfusion, transfusion, traumatic brain injury, inflammation, and clinical outcomes.
