  • 4741.识别肿瘤细胞为基础的DNA损伤反应微环境导致癌症治疗抵抗

    A major impediment to effective prostate cancer treatment involves the acquired resistance to cytotoxic therapies. Components of the tissue microenvironment are increasingly recognized to profoundly influence tumor cell phenotypes that include susceptibilities to toxic insults. Using a genome-wide analysis of transcriptional responses to genotoxic stress induced by cancer therapeutics, we have identified a spectrum of secreted proteins derived from the tumor microenvironment (TME) that have the potential to modify tumor growth and enhance resistance to DNA-damaging cancer therapeutics. These results suggest a mechanism by which genotoxic therapies given in a cyclical fashion can enhance subsequent treatment resistance through cell non-autonomous effects contributed by the TME. To date, the contributions of individual members of this DNA Damage-associated Secretory Program (DDSP) have not been defined, nor have the signaling mechanisms responsible for propagating the DNA-damage signal(s) been determined. Our objective during this grant period is to test whether treatment-associated DNA damage responses in cells comprising the prostate TME promote tumor growth and subsequent therapy resistance. During this funding period we have: (1) Generated a prostate fibroblast cell line stably expressing SPINK1; (2) Evaluated the impact which SPINK1 activation has upon the growth characteristics of prostate cancer cells lines; (3) Examined how SPINK1 secretion from the microenvironment modulates the response of prostate cancer cells to chemotherapeutics: (4) Begun the evaluation of SPINK1 regulatory pathway.

    In the current monitoring system, for the monitoring sites (such as storage, home, radioactive places and other harmful places), no dead ends and comprehensive monitoring is completed by installing a large number of fixed cameras and hiring many security persons to achieve, a lot of cameras installed are complicated; expensive and wasting a lot of manpower and material resources. Meanwhile, the fixed camera monitoring is just rigid; no early warning functions and requiring the security persons to be on duty all the time. At the same time, the records of on-site monitoring need to be large capacity storage medium. Therefore, to solve the problems of above in practical, we design an intelligent network monitoring system which is a set of non-dead mobile monitoring; automatic warning; automatic voice alerting; automatic storage of suspicious scene. By fixing the cameras; wireless routers; GPRS module; voice module and power management modules, with the Altera DE2 board to build a wheeled robot. The wheeled robot realizes the real-time monitoring of unmanned sites and solves the issues of complex installing and adverse monitoring to a large extent.

    Thermal energy storage in buildings dates back to the earliest human settlements. Houses built by indigenous people all across the world, in hot and moderate climates, demonstrate some form of thermal energy storage. The most common and easiest form of thermal energy storage uses the diurnal temperature change. Diurnal temperature difference is the difference between the minimum at night, usually lower, and the maximum during the day, usually higher. This leads to colder nights and warmer days. The colder nights provide a "night cool," which can be used to cool the hot days, if the energy can be stored and released. The opposite is also possible—storing the warmth of the day to heat the nights. The indigenous homes stored the night cool in the mass of walls, floors, and roofs. This method can be used in modern and present-day buildings.

    This technical report is duplicative documentation of the approved doctorial thesis of one of the co-authors. The research was funded in part by the U.S. Army Aviation and Missile Research, Development, and Engineering Center (AMRDEC) for the development of micromechanical sensors under the Army Technology Objective (ATO) of Sensor, Warhead, and Fuzing Technology Integrated for Combined Effects (SWFTICE). Particular technical progress at AMRDEC within this report includes resonant array processing (Chapter 3), electrets integration with Microelectromechanical Systems (MEMS) with localized heater fabrication for wafer bonding and microcharging grids for in-situ charging using microcoronas (Chapter 4), and processing of MEMS transducers (Chapter 5).

    The color and shape feature are very important quality characteristic for classification of fruits. The dominant grading color histogram feature and radius normal angle histogram feature were presented in this paper. They can well represent the color and shape information of fruits respectively and are not sensitive to the changes of scale, translation and rotation. Experiment results showed both histogram features can effectively distinguish between different grade fruits and have high classification accuracy. They are suitable for real-time application.

    This report describes an algorithm, implemented in Matlab/Simulink, designed to reduce the variability of photovoltaic (PV) power output by using a battery. The purpose of the battery is to add power to the PV output (or subtract) to smooth out the high frequency components of the PV power that that occur during periods with transient cloud shadows on the PV array. The control system is challenged with the task of reducing short-term PV output variability while avoiding overworking the battery both in terms of capacity and ramp capability. The algorithm proposed by Sandia is purposely very simple to facilitate implementation in a real-time controller. The control structure has two additional inputs to which the battery can respond. For example, the battery could respond to PV variability, load variability or area control error (ACE) or a combination of the three.

    The integration and stability of electrocatalytic nanostructures, which represent one level of porosity in a hierarchical structural scheme when combined with a three-dimensional support scaffold, has been studied using a combination of synthetic processes, characterization techniques, and computational methods. Dendritic platinum nanostructures have been covalently linked to common electrode surfaces using a newly developed chemical route; a chemical route equally applicable to a range of metals, oxides, and semiconductive materials. Characterization of the resulting bound nanostructure system confirms successful binding, while electrochemistry and microscopy demonstrate the viability of these electroactive particles. Scanning tunneling microscopy has been used to image and validate the short-term stability of several electrode-bound platinum dendritic sheet structures toward Oswald ripening. Kinetic Monte Carlo methods have been applied to develop an understanding of the stability of the basic nano-scale porous platinum sheets as they transform from an initial dendrite to hole containing sheets. Alternate synthetic strategies were pursued to grow dendritic platinum structures directly onto subunits (graphitic particles) of the electrode scaffold. A two-step photocatalytic seeding process proved successful at generating desirable nano-scale porous structures. Growth in-place is an alternate strategy to the covalent linking of the electrocatalytic nanostructures.

    The Texas Panhandle is regarded as the Cattle Feeding Capital of the World, producing 42of the fed beef cattle in the United States within a 200-mile radius of Amarillo generating more than 5 million tons of feedlot manure /year. Apart from feedlots, the Bosque River Region in Erath County, just north of Waco, Texas with about 110,000 dairy cattle in over 250 dairies, produces 1.8 million tons of manure biomass (excreted plus bedding) per year. While the feedlot manure has been used extensively for irrigated and dry land crop production, most dairies, as well as other concentrated animal feeding operations (CAFOs), the dairy farms utilize large lagoon areas to store wet animal biomass. Water runoff from these lagoons has been held responsible for the increased concentration of phosphorus and other contaminates in the Bosque River which drains into Lake Wacothe primary source of potable water for Wacos 108,500 people.

    Despite the elevated rates of psychiatric problems among returning combat veterans, available evidence suggests that as many as half of soldiers screening positive for mental health problems never seek treatment for these issues (Fikretoglu et al., 2008, Hoge et al., 2006). One promising treatment approach that has shown efficacy in preliminary research and which may address issues related to stigma and barriers to care, is the used of web-based treatment interventions. In particular, internet-based cognitive behavioral therapy (iCBT) is rapidly emerging as a potentially efficacious treatment option for many individuals with mild to moderate depression (Andersson and Cuijpers, 2009). Emerging evidence suggests that iCBT is a particularly promising and well-accepted approach for treating large numbers of individuals while minimizing cost and clinicians time demand. Recently, researchers from the School of Psychiatry at the University of New South Wales (UNSW) developed and validated several, clinician-assisted iCBT programs that have shown remarkable success in treating major depressive disorder (MDD), generalized anxiety disorder, social phobia, and panic disorder (Robinson et al., 2010, Titov et al., 2010, Titov et al., 2009).

    Tamoxifen, the first targeted therapy, has shown great success in treating estrogen receptor (ER) positive breast cancer. However, both acquired and de novo resistance to this therapy prevents it from being effective in all situations. Multiple lines of evidence indicate that increased signaling through growth factor pathways, such as the IGF pathway, mediates resistance to tamoxifen. The link between ER and ICGF1R leads us to hypothesize that IGF system crosstalk with the ER contributes to tamoxifen resistance. Tamoxifen resistance has thus provided researchers with a reason to investigate other growth factor pathways involved in breast cancer. As new targeted therapies are being developed, it will be important to examine their benefit with existing therapies. In order to examine the effectiveness of anti-IGF1R inhibitors in vitro, tamoxifen resistant (TamR) cells were generated by culturing MCF-7L and T47D cells in the presence of 4-OH-tamoxifen for >6 months. TamR cells had dimished levels of IGF1R, with unchanged levels of insulin receptor (IR). Further, TamR cells failed to respond to IGF-I induced p-AKT activation, while retaining responsiveness to both insulin and IGF-II. Additionally, IGF-I failed to enhance the proliferation and anchorage-independent growth of TamR cells; however, both insulin and IGF-II were able to enhance proliferation in MCF
    anchorage-independent growth. An IGF1R antibody was effective in inhibiting signaling, anchorage-independent growth, and proliferation in MCF-7L cells, but had no effect in TamR cells. In contract, an IGF1R/TR tyrosine kinase inhibitor was effective in both MCF-7L and TamR cells. In a xenograft model, an IGF1R antibody was able to inhibit estrogen stimulated tumor growth, but had no additive effect when combined with tamoxifen treatment. Further, tamoxifen-treated xenografts had diminished IGF1R levels.
