2871.用于肿瘤脑膜炎治疗的病毒溶瘤细胞

Neoplastic meningitis is a severe complication of breast cancer for which there is no curative treatment at present. The project aims to develop a novel, safe and efficient therapy for neoplastic meningitis HSV-1 oncolysis therapy. For the first year of the award period we have propagated the HSV-1 replication conditional mutant (hrR3) with burst assays and generated three different virus preparations to establish viral titers of 1x10(7), 1x10(8) and 1x10(9) pfu/ml. The virus titers were determined by plaque assays performed on vero mono-layers. These virus titers will be used to study fractionated viruses particles. In addition we have commenced on additional work that will help study the disease progression by molecular imaging. In anticipation of establishing a mouse model of meningeal metastasis we have developed stable breast cancer cell lines that express bioluminescence and fluorescence markers for in vivo molecular imaging. Three cell lines (human breast cancer cell lines SkBr3 and MDA-231, and the mouse breast cancer 4T1) were transfected to express renilla luciferase (RLuc) and mCherry, both driven by the CMV promoter. Clones that were resistant to puromycin were selected to establish stable cell lines. These stable cell lines have been tested for (i) the replication efficiency HSV- 1, and (ii) their sensitivity to the HSV-1 oncolysis with in vitro bioluminescence. Based on these preliminary data we anticipate proposing the use of one of the cell lines to establish a mouse model of meningeal metastases. We will in due course approach DOD for approval of the animal protocol.

2872.统一性创作工具的异构设备和协议

A contents authoring software with device management for interactive shows and performances is proposed. The proposed system allows user to register and to manage various stagerelated devices such as lighting system, fog generator, robots or multimedia players. In this paper, we describe software architecture, XML specification for unified protocol description, and user interface of the proposed system. The system can control multiple devices of heterogeneous protocols in timely manner. Once devices are registered, user can browse commands of each device and can organize commands to stepbased list according to the performance scenario. Unlike current commercialized products, the system allows to manipulate authored list of commands during the show instantly.The proposed system also supports to control playback of authored commands by sensor inputs so that a player can trigger devices to execute according to events. The contributions of this paper can be summarized to three points; (1) define unified protocol description with XML structures for simplicity (2) give sensor triggering capability to control devices, and (3) implement fast and instant manipulation user interface during the show.

To conclude this monograph, we want to recall the general philosophy behind the approach developed from chapter to chapter as well as the difficulty we can encounter when designing a structured controller and thus the development that needs to be done. As a matter of fact there are three major concepts that have to be considered: the concept of system, and the concept of control intended to transform the original one into another with a more satisfactory behavior. This transformation is made through the use of a design procedure, which is the third concept we want to talk about.

2874.生物可信视觉伺服控制的实施和验证

In this report, we compare the performance of the Lucas-Kanade algorithm with feature tracking with the bioplausible optical flow algorithm to achieve pose stabilization in the context of one-dimensional (1-D) attitude stabilization. These results have been benchmarked against an ideal controller in both simulation and robotic experimentation. Within both environments, the accuracy, stability, and settling time were evaluated as a function of contrast and system noise. Both algorithms achieved reasonable performance when compared to the ideal controller, but in most cases the Lucas-Kanade algorithm outperformed the bioplausible algorithm. Within the evaluated serial implementation, the Lucas-Kanade algorithm was also faster. It is anticipated that both performance and processing speed will improve for the bioplausible algorithm when it is implemented in a parallel instantiation.

Timc-varying linear quadratic Gaussian (LQG) control for vibration of coupled vehicle-bridge system is studied.The vehicle is modeled as a moving mass model with three degrees of freedom,which consists of vehicle body,bogie and wheel.Active suspensions are adopted for the primary and secondary ones,and the control forces are produced by two actuators placed between the bogie and wheel,and between the vehicle body and the bogie,respectively.Vehicle-bridge coupling systems are time-dependent,which lead to the time-varying Riccati differential equation and the time-varying Kalman-Bucy filter equation in the LQG controller design.However,both of them are solved precisely via precise integration method and symplectic conservative perturbation method.In the example,the time history responses of the bridge and the vehicle were calculated respectively for the vehicle with passive suspensions or with active suspensions.Numerical results show that with active suspensions adopted,ride comfort can be improved when the vehicles passing through the bridge.

2876.乳腺癌的非热能高强度聚焦超声治疗

Several in vitro studies have demonstrated the non-thermal (< 42 deg C) cell killing effect of pulsed HIFU, which resembles high linear energy transfer (LET) radiation cell damage that is not not affected by the local biochemical environment and shows less radiation resistance. However, there have been no in vivo animal studies performed on non-thermal HIFU using an animal model by comparing it with well-established treatment modalities such as radiation therapy. This project has two aims: (1) to determine if non-thermal HIFU can cause significant tumor growth delays in vivo as compared to radiotherapy using an animal model; and (2) to determine potential normal tissue toxicities, if any, associated with non-thermal HIFU treatment for breast cancer. Extensive phantom studies have been completed to determine suitable ultrasound parameters for the in vivo animal experiments demonstrated significant tumor growth delay by non-thermal HIFU that are comparable to radiotherapy without damage to overlapping tissues, which warrant future feasibility studies to determine optimal HIFU treatment parameters for human treatments and to quantify normal tissue toxicities associated with non-thermal HIFU treatments.

2877.全向康复训练沃克的健壮容错跟踪控制

This paper is concerned with the problem of robust fault-tolerance tracking control on the omni-directional rehabilitative training walker. The nonlinear robust fault-tolerance method is proposed when one wheel actuator stuck is happened. The aim is to design an asymptotically stable controller which can guarantee the safety of walker and can ensure tracking the training path planned by a physical therapist when one actuator is faulted. The controller which can satisfy asymptotic stability is obtained by constructing the energy storage function and the γ dissipation inequality. The evaluation signal is designed which has verified the nonlinear L_2 gain performance index. Finally, the simulation results confirm the feasibility and effectiveness of the designed scheme.

2878.卵巢癌醛脱氢酶细胞群的描述和定位

Despite a common outstanding response to primary therapy, most ovarian cancer patients will experience recurrence due to what is often microscopic undetectable disease. One possible cause of this is a chemoresistant population of cells with stem cell characteristics. We have examined one potential population in particular, the ALDH-positive population. We have shown that ALDH1A1-positive cells are more tumorigenic than ALDH1A1- negative cells, contribute to poor patient outcomes, and contribute to chemoresistance. Importantly, these effects can be reversed by downregulating ALDH1A1 expression with nanoparticle-delivered siRNA. Additionally, we have shown that CSCs are clinically significant, in that chemoresistant tumors have increased density of ALDH and CD133 cells. Thus they likely represent at least part of the chemoresistant population within a heterogeneous tumor. Importantly, they do not seem to explain the entire story, as there are still many CSC-negative cells present at the conclusion of treatment. Additional studies will be performed to determine which other cell types may be present in chemoresistant tumors, and which pathways or mechanisms may be mediating this resistance.

A systematic review that included 110 reports of eligible studies published from January 1985 through January 2011 was undertaken to determine the potential benefits and adverse effects of screening, monitoring, and treatments for adults with chronic kidney disease (CKD) stages 13. CKD stages 13 are defined as: 1 (kidney damage with a glomerular filtration rate (GFR) >90 mL/min/1.73 m2), 2 (kidney damage with a GFR of 6089 mL/min/1.73 m2), or 3 (a GFR of 3059 mL/min/1.73 m2, regardless of kidney damage). The systematic review excluded studies of patients with CKD stages 4 and 5. This is a summary of the systematic review meant to assist in decisionmaking along with a patients values and preferences and should not be construed to represent clinical recommendations or guidelines.

This document specifies the data interface for TSAFE, the Tactical Separation-Assured Flight Environment. TSAFE is a research prototype of a software application program for alerting air traffic controllers to imminent conflicts in enroute airspace. It is intended for Air Route Traffic Control Centers ('Centers') in the U.S. National Airspace System. It predicts trajectories for approximately 3 minutes into the future, searches for conflicts, and sends data about predicted conflicts to the client, which uses the data to alert an air traffic controller of conflicts. TSAFE itself does not provide a graphical user interface.
