A major impediment to effective prostate cancer treatment involves the acquired resistance to cytotoxic therapies. Components of the tissue microenvironment are increasingly recognized to profoundly influence tumor cell phenotypes that include susceptibilities to toxic insults. Using a genome-wide analysis of transcriptional responses to genotoxic stress induced by cancer therapeutics, we have identified a spectrum of secreted proteins derived from the tumor microenvironment (TME) that have the potential to modify tumor growth and enhance resistance to DNA-damaging cancer therapeutics. These results suggest a mechanism by which genotoxic therapies given in a cyclical fashion can enhance subsequent treatment resistance through cell non-autonomous effects contributed by the TME. To date, the contributions of individual members of this DNA Damage-associated Secretory Program (DDSP) have not been defined, nor have the signaling mechanisms responsible for propagating the DNA-damage signal(s) been determined. Our objective during this grant period is to test whether treatment-associated DNA damage responses in cells comprising the prostate TME promote tumor growth and subsequent therapy resistance. During this funding period we have: (1) Generated a prostate fibroblast cell line stably expressing SPINK1; (2) Evaluated the impact which SPINK1 activation has upon the growth characteristics of prostate cancer cells lines; (3) Examined how SPINK1 secretion from the microenvironment modulates the response of prostate cancer cells to chemotherapeutics: (4) Begun the evaluation of SPINK1 regulatory pathway.

The complete annual report for 1977 gives information on supported projects in the Program for Energy Research and Technologies, 1977--1980 of the Federal Government excluding projects relating to nuclear energy and fusion research; it is broken down into three subprograms: (1) efficient use of energy in application and in secondary energy; (2) coal and other fossil sources of primary energy; (3) new sources of energy. This document presents only subprogram (3), new sources of energy, as excerpted from the overall annual report, namely: development of new energy sources (solar energy, wind energy, geothermal energy) through ascertaining energy availability and its regional distribution; development of technologies for use and conversion; and demonstration of technical and economic feasibility. Ninety-four projects are described, including objectives, funding, and status. Additional information on the projects is included in appendix.

Despite intense research efforts, cancer remains the second leading cause of death in the United States. Mortality is seldom caused by primary tumors, but rather by the effect of metastases on distant organs. The lymphatic system serves as a common route of metastasis for many cancers of epithelial origin. There is growing evidence that lymphangiogenesis, the sprouting of new lymphatics from pre-existing lymphatics, facilitates the dissemination of cancer. Interestingly, vascular endothelial growth factor receptor-2 (VEGFR2) signaling has been shown to stimulate lymphangiogenesis in adult mice. However, the role VEGFR2 serves in the development of the lymphatic system has not been defined. Here we use the Cre-lox system to show that the proper development of the lymphatic vasculature requires VEGFR2 expression by lymphatic endothelium. This newly identified function of VEGFR2 further defines the molecular pathways controlling the development of the lymphatic vasculature and sheds light on how therapeutic agents targeting VEGFR2 can inhibit tumor lymphangiogenesis.

Rich client-side applications written in HTML5 proliferate diverse platforms such as mobile devices, commodity PCs, and the web platform. These client-side HTML5 applications are increasingly accessing sensitive data, including users’ personal and social data, sensor data, and capability-bearing tokens. To fulfill their security and privacy guarantees, these applications need to maintain certain data-confinement invariants. These invariants are not explicitly stated in today’s HTML5 applications and are enforced using ad-hoc mechanisms. The complexity of web applications,coupled with hard-to-analyze client-side languages, leads to low-assurance data-confinement mechanisms in which the whole application needs to be in the TCB to ensure data-confinement invariants.

The VA Office of Inspector General (OIG) Office of Healthcare Inspections conducted a review in follow up of its March 2011 report on radiation therapy (RT) at the VA Long Beach Healthcare System (facility) in Long Beach, CA. OIG also assessed the validity of new allegations related to the quality of radiation treatments. We conducted an unannounced inspection in December 2012 and an announced inspection in February 2013, observed the practices of radiation therapists, and examined departmental electronic and paper records that are accessible only onsite.

In 2008, wind energy enjoyed another record-breaking year of industry growth. By installing 8,358 megawatts (MW) of new generation during the year, the U.S. wind energy industry took the lead in global installed wind energy capacity with a total of 25,170 MW. According to initial estimates, the new wind projects completed in 2008 account for about 40of all new U.S. power-producing capacity added last year. The wind energy industrys rapid expansion in 2008 demonstrates the potential for wind energy to play a major role in supplying our nation with clean, inexhaustible, domestically produced energy while bolstering our nations economy. To explore the possibilities of increasing winds role in our national energy mix, government and industry representatives formed a collaborative to evaluate a scenario in which wind energy supplies 20of U.S. electricity by 2030. In July 2008, the U.S. Department of Energy (DOE) published the results of that evaluation in a report entitled 20Wind Energy by 2030: Increasing Wind Energys Contribution to U.S. Electricity Supply. According to the report, the United States has more than 8,000 gigawatts (GW ) of available land-based wind resources that could be captured economically. In the early release of its Annual Energy Outlook 2009, the U.S. Energy Information Administration (EIA) estimates that U.S. electricity consumption will grow from 3,903 billion kilowatt-hours (kWh) in 2007 to 4,902 billion kWh in 2030, increasing at an average annual rate of 1. To meet 20of that demand, U.S. wind power capacity would have to reach more than 300 GW (300,000 MW). This growth represents an increase of more than 275 GW within 21 years. Although achieving 20wind energy will have significant economic, environmental, and energy security benefits, to make it happen the industry must overcome significant challenges.
