Background. For diagnostic processes involving continual measurements from a single patient, conventional test characteristics, such as sensitivity and specificity, do not consider decision consistency, which might be a distinct, clinically relevant test.

Control murine liver endothelial cells and murine breast tumor endothelial cells were successfully isolated and plated for the imaging and flow cytometry experiments. I have found interesting differences between normal and tumor endothelial cells well worth further studying. More specifically, I have found that the response of tumor endothelial cells to VEGF is the same regardless of whether or not they have been serum-starved. Also receptor densities of VEGFR-1 and -2 differ between normal and breast tumor ECs.

We have successfully prepared sense expression libraries, characterized the size of their inserts which are fused to a green fluorescent protein tag, and packaged these libraries into lentivirus. Four different human MDS bone marrow isolates have been transfected with lentivirus at multiplicity of infection of 0.1. A collection of 252 puromycin resistant clones were isolated, genomic DNA was prepared, and clones were screened for ability to undergo myeloid differentiation in response to GM-CSF. Only approximately 30% of the puromycin resistant clones (78) acquired this phenotype. Transfection of MDS cells with antisense libraries did not generate any clones that acquired the desired (differentiative) phenotype. PCR was used to identify presence of the trans gene in the 78 clones. DNA sequencing has identified the cDNA inserts. These inserts were expressed in na ve MDS cell pools. Eight of these cDNAs were validated; they induced myeloid colonies in vitro and engrafted in the marrow of SG3, but not NSG mice.

The aim of this study was to characterize the type, topography and severity of developmental changes in the brain of people diagnosed with autism. To detect global pattern of brain pathology, the entire brain hemisphere was embedded in celloidin or polyethylene glycol and cut into hemispheric sections. Examination of CV stained serial sections revealed a broad spectrum of brain structure changes (defects of neurogenesis, migration, gyrification and dysplasia) affecting both gray and white matter in the brain and cerebellum of autistic subjects including: subependymal nodular dysplasia and thickening of subependymal layer; polymicrogyria; cerebellar hypoplasia and floculonodular dysplasia; broad spectrum of dysplastic changes in the neo- and archicortex, cornu Ammonis, dentate gyrus and amygdala; and heterotopias in the white matter in cerebellum, brain hemisphere and hippocampus. The study of 22 brains of subjects with idiopathic autism revealed changes in approximately 90% of cases and in all four examined brains of subjects with autism/Ch15Dupl. The type of detected changes indicates that all of these changes reflect fetal developmental alterations. Presence of these changes in autistic subjects, regardless of age, indicates that they are permanent alterations of brain structure and may contribute to the core clinical phenotype of autism.

Physicians often lack the time to familiarize themselves with the details of particular allergies or other drug restrictions. Clinical Decision Support (CDS), based on a structured terminology as SNOMED CT (SCT), can help physicians get an overview, by automatically alerting allergy, interactions and other important information. The centralized CDS platform based on SCT, controls Allergy, Interactions, Risk Situation Drugs and Max Dose restrictions by the help of databases developed for these specific purposes. The CDS will respond to automatic web service requests from the hospital or GP electronic medication system (EMS) during prescription, and return alerts and information. The CDS also contains a Physicians Preference Database where the physicians individually can set which kind of alerts they want to see. The result is clinically useful information physicians can use as a base for a more effective and safer treatment, without developing alert fatigue.
