251.p53基因突变W248和H175对MCF-7细胞的生物学效应

Objective:p53,a tumor suppressor gene,is one of the hotspots in the world of the biomedical field.Mutation of p53 gene,which is found in approximately 50% of human cancers,is a key event in carcinogenesis.This project aims to investigate the new characteristics of two p53 mutants,p53-W248 and p53-H175,in MCF-7 cells,so as to provide the experimental basis for understanding the functional alternations of mutant p53.Methods:In this study,MCF-7 cells transfected with p53-H175 or p53-W248 plasmids were used as experimental group and the MCF-7 cells transfected wild type p53 plasmid were used as control group.Then the biological effects at the cellular level were investigated using 3-(4.5-dimethylthiazol-2-yl)-2,5-diphenyl tetrazolium bromide (MTT) assay,flow cytometry analysis and cell scratch test.Results:MTT assay showed that p53-W248 might promote cell proliferation in MCF-7 cells.The results of flow cytometry indicated that no significant effect on cell cycle progression and cell apoptosis by p53-H175 or p53-W248 in cells.The cell scratch test showed that p53-H175 could increase the ability of cell migration.Conclusion:p53-H175 could lead to the promotion of tumor cell migration,while p53-W248 may promote tumor cell proliferation,p53-H175 and p53-W248 might have acquired some new characteristics of oncogenes.

NREL and Solar Power Industries will cooperate to evaluate technology for producing solar grade silicon from industrial waste of the phosphorus industry, as developed by the Institute of Physics and Technology (IPT), Kazakhstan. Evaluation will have a technical component to assess the material quality and a business component to assess the economics of the IPT process. The total amount of silicon produced by IPT is expected to be quite limited (50 kg), so evaluations will need to be done on relatively small quantities (5 kg/sample). Department of Energy (DOE) laboratories, including NREL, have been placing subcontracts for research and development services with scientific institutes in the Newly Independent States (NIS) under various DOE programs for many years.

At the request of the USAF Radioisotope Committee Secretariat (RICS), the USAF School of Aerospace Medicine Consultative Services Division completed an independent radiological assessment/verification survey of sites 321 B North and PRL-S006 at former McClellan AFB, CA, from 12-13 Sep 12. This verification survey was conducted at each of the sites post-remediation and after the contractor had completed its Final Status Survey (FSS) sampling. Radium-226 was the sole radionuclide of concern. Environmental Dimensions, Inc., under contract with CH2M Hill, conducted all radiological field work to include FSSs. This letter details the findings of this visit and is meant to assist the RICS when evaluating the contractor’s FSS reports of these sites.

Purpose: To develop a robust and rapid imaging technique for hyperpolarized 13C MR Spectroscopic Imaging (MRSI) and investigate its performance. Methods: A concentric rings readout trajectory with constant angular velocity is proposed for hyperpolarized 13C spectroscopic imaging and its properties are analyzed. Quantitative analysis of design tradeoffs are presented for several imaging scenarios in terms of resolution, spectral bandwidth, acquisition time and SNR efficiency. Analysis comparing parallel imaging accelerated Cartesian sampling and concentric rings in terms of reconstruction noise amplification are also presented. Finally, the first application on 13C phantoms and in vivo animal hyperpolarized 13C MRSI studies were performed to demonstrate the feasibility of the proposed method. results: The concentric rings MRSI trajectory has the advantages of acquisition timesaving compared to echo-planar spectroscopic imaging (EPSI), and robustness to system timing delay and eddy currents compared to spiral MRSI. It provides a sufficient spectral bandwidth with a relatively high SNR efficiency compared to EPSI and spiral techniques. It will also be beneficial for parallel imaging by reducing noise amplificiation. Conclusion: The concentric rings trajectory is a robust and rapid imaging technique that fits very well with the speed, bandwidth, and resolution requirements of hyperpolarized 13C MRSI.

Classification using portable advanced electromagnetic sensors, designed to operate in areas where terrain and vegetation preclude the use of vehicle-borne sensors, was demonstrated at the former Camp Beale, CA in 2011. The TEMTADS 2x2, Man-Portable Vector (MPV) and portable Berkeley UXO Discriminator (BUD) were used to successfully classify all of the targets of interest by all analysts. There was some variation among analysts in the percent of clutter rejected. All but two of the 16 performers eliminated about 75of the clutter. This is comparable to the results achieved using the vehicular-borne MetalMapper on another part of this site. Even though this was the first live site demonstration for each of the sensors, daily productivity of 90 to 175 anomalies was possible. One of the teams was able to collect cued data, extract parameters, and classify for $26 per anomaly. Using this per anomaly cost for classification and a few reasonable assumptions we calculate that the use of classification would result in a 50savings for a 100-acre remediation on a site with conditions like Camp Beale. When a site is remediated, it is typically mapped with a geophysical system, based on either a magnetometer or electromagnetic induction (EMI) sensor, and the locations of all detectable signals are excavated. Many of these detections do not correspond to munitions, but rather to other harmless metallic objects or geology: field experience indicates that often in excess of 99of objects excavated during the course of a munitions response are found to be nonhazardous items. As a result, most of the costs to remediate a munitions- contaminated site are currently spent on excavating targets that pose no threat. If these items could be determined with high confidence to be nonhazardous, some of this expense could be avoided and the available funding applied to more sites.

Summary: Proof-of-concept of Artificial Hair Sensor with CNT mechano- resistive elements demonstrated. CNT arrays examined that initially appear mechanically different by length, but DIC reveal a common failure criteria. Observed carbon hair deflection in-phase with gust-like disturbances without vibration - mapped the flow regime steady/unsteady. Computed frequency response plots for carbon hair structure in quasi-steady regime 2nd order like response. Found that elasticity and drag dominated the force balance in the carbon hair structure. Established criteria based on dimensionless groups to control hair dynamic response through selection of material and geometric properties.

The Idaho National Laboratory (INL) has initiated an effort to evaluate the viability of using ultrasonic thermometry technology as an improved sensor for detecting temperature during irradiation testing of advanced fuels proposed within the Fuel Cycle Research and Development (FCR&D) program sponsored by the U.S. Department of Energy (US DOE). Ultrasonic thermometers (UTs) work on the principle that the speed at which sound travels through a material (acoustic velocity) is dependent on the temperature of the material. UTs have several advantages over other types of temperature sensors . UTs can be made very small, as the sensor consists only of a small diameter rod which may or may not require a sheath. Measurements may be made up to very high temperature (near the melting point of the sensor material) and, as no electrical insulation is required, shunting effects observed in traditional high temperature thermocouple applications are avoided. Most attractive, however, is the ability to introduce multiple acoustic discontinuities into the sensor, as this enables temperature profiling with a single sensor. The current paper presents initial results from FCR&D UT development efforts. These developments include improved methods for fabricating magnetostrictive transducers and joining them to waveguides, characterization of candidate sensor materials appropriate for use in FCR&D fuels irradiations (both ceramic fuels in inert gas and sodium bonded metallic fuels), enhanced signal processing techniques, and tests to determine potential accuracy and resolution.

Fish passage conditions through a Kaplan turbine and spillway fish weir at Foster Dam, located on the South Santiam River in Linn County, Oregon, were evaluated by Pacific Northwest National Laboratory for the U.S. Army Corps of Engineers, Portland District, using Sensor Fish devices. The objective of the study was to describe and compare passage exposure conditions, identifying potential fish injury regions encountered during passage via specific routes. The investigation was performed in May 2012, concurrent with HI-Z balloon-tag studies by Normandeau Associates, Inc. Sensor Fish data were analyzed to characterize the passage exposure conditions through the spillway fish weir and turbine Unit 1 at Foster Dam at two forebay pool elevations (616 and 634 ft mean sea level (MSL)) and to estimate data relationships with live fish injury and mortality estimates. For the spillway fish weir evaluation, Sensor Fish and live fish were deployed through injection system piping mounted on the weir. The bottom of the injection pipe was at an elevation of approximately 614 ft during testing at the 616-ft MSL (low) forebay elevation and at 632 ft during the 634-ft MSL (high) forebay level tests. Two systems were useda 4-in. pipe for juvenile fish releases and an 8-in. pipe for adult releases at each elevation.

A multiyear pilot energy efficiency retrofit project has been undertaken by Greenbelt Homes, Inc, (GHI) a 1,566 co-operative of circa 1930 and 1940 homes in Greenbelt, Maryland. The three predominant construction methods of the townhomes in the community are materials common to the area and climate zone including 8 in. concrete masonry unit (CMU) block, wood frame with brick veneer and wood frame with vinyl siding. GHI has established a pilot project that will serve as a basis for decision making for the rollout of a decade-long community upgrade program that will incorporate energy efficiency upgrades to the building envelope from the exterior and equipment with the modernization of other systems like plumbing, mechanical equipment, and cladding. The pilot project has three phases focused on identifying the added costs and energy savings benefits of energy efficiency features that are to be installed during a planned community-wide replacement program commencing in 2015.
